Abstract

Calcification-related complications are the primary cause of morbidity and mortality in CKD patients. The serum calcification propensity predicts their survival. Calciprotein particles (CPP) are thought to be responsible for the chronic inflammation in CKD patients; however, the regulation of the formation and stability of CPP are insufficiently characterized. We hypothesize that a humoral system prevents calcification in serum, and impaired control of this system causes extraosseous calcifications, inflammation and oxidative stress. We propose to characterize the composition and properties of model CPP and CPP from hemodialysis and CKD patients, and investigate the biological responses of cells exposed to CPP. The proposed research has the potential to identify novel calcification inhibitors/promoters, which may improve preventative care and lead to novel therapies for CKD patients.
